    Study Notes

    Lab Procedures: Differential Tests 2

    • Work as a table group
    • Label all tubes/plates/racks
    • Do not tighten caps on inoculated tubes

    Labeling Tubes

    • Use white masking tape to label each tube
    • Include table symbol, section time, test abbreviation, species name
    • Example: MW11:50 MRVP E.coli

    Today's Inoculation (Additional Notes)

    • Work as a table
    • Label tubes, plates, and racks
    • Leave caps loose on inoculated tubes
    • Another group member will pick up the Gelatin hydrolysis stab from the table and incubate at room temperature.
    • Combine all tubes into one rack and label
    • Tape together five plates and label
    • Place inoculated materials into the 37°C incubator for 16 hours.

    Biochemical Tests (2nd page)

    • Bacterial identification needs to utilize multiple tests
    • Includes gelatin hydrolysis stab, catalase tests, nitrate reduction, DNA hydrolysis, lipid hydrolysis, blood agar, oxidase tests, and others (as listed in the document).

    Unknown Pathogenic Bacteria (3rd page)

    • Growth considerations during isolation & identification: media type, nutrients, incubation temperature, oxygen conditions, pH, salt.
    • Isolate colonies as pure cultures
    • Identify genus & species of microorganisms

Identification of Physical Characteristics (3rd page)

    • Microorganisms are often identified by physical characteristics like shape and gram reactions before using more complex biochemical tests.
    • Various staining techniques are used to identify bacterial morphology:
      • Simple Stain (cell shape)
      • Gram Stain (peptidoglycan content, gram + or gram -)
      • Capsule Stain (capsule presence/absence)
      • Endospore Stain (endospore presence/absence)
      • Acid Fast Stain (presence of mycolic acid).

    Identification, Biochemical Characteristics (4th page)

    • Utilize biochemical tests to determine if the tested organisms have the appropriate enzymes to metabolize specific molecules
    • Tests to look for are:
      • Degradation enzymes (Catalase, Urease, Lipase).
      • Sugar fermentation (Mannitol, Lactose, Glucose)
      • Nitrate reductase
      • Tryptophanase
      • Phenylalanine deaminase
      • Citrate as sole carbon source
      • Malonate as sole carbon source
      • Fermentation end products (Methyl red, Voges-Proskauer)
    • Results of tests are analyzed to determine the identity of the unknown organism

Results of Biochemical RXN (4th page)

    • Results are recorded and used to aid in the identification process
    • Examples of tests used include catalase (POS), urea (NEG), phenol red lactose (POS), and others.
    • Interpretation is key to determining usefulness of a test to further identify the organism.

    Bergey's Manual (5th page)

    • Use the "Determinative" edition, not the systematic one
    • Complete data tables first.
    • Create flowcharts using PowerPoint
    • Include a minimum of 7 tests for flowcharts.
    • Submit flowcharts and tables properly for grading purposes.
