Blut und Blutbestandteile
- Blut besteht aus Erythrozyten (Rothemoglobin), Leukozyten (weiße Blutkörperchen) und Thrombozyten (Blutplättchen)
- Blutplasma: 90% Wasser, 10% gelöste Bestandteile (Proteine, Elektrolyte, Hormone, Nahrungsstoffe)
- Blutserum + Fibrinogen = Blutplasma
Funktionen des Blutes
- Transport und Kommunikation (Nahrungsstoffe, Sauerstoff, Metabolite, Abfallstoffe, Elektrolyte, Hormone)
- Pufferfunktion: Konstanterhaltung des pH-Werts
- Abwehr und Schutz
- Hämostase: Blutstillung und Blutgerinnung
- Regulationsfunktion (Temperatur, Wasserhaushalt)
Aminosäuren
- AS sind in Lösung bei neutralem pH-Wert vorwiegend ionisiert
- Dipolares Zwitterion: Aminogruppe ist protoniert (NH3+), Carboxylgruppe ist dissoziiert/deprotoniert (COO-)
- AS sind Ampholyte und wirken als Puffer
- Klassifizierung der 21 proteinogenen AS in 4 Gruppen (hydrophob, polare, positiv geladene, negativ geladene)
Proteine
- Membranproteine: interagieren mit hydrophober Lipidschicht, ca. 30% aller Proteine
- Typen von Membranproteinen:
- Typ 1 und 2: nur eine Transmembranhelix, unterschiedliche Orientierung der N- und C-Termini
- Typ III: mehrere Helices, 7 Rezeptoren (z.B. beta-adrenerge Rezeptoren)
- Typ IV: mehrere Membranproteine lagern sich zu Quartärstruktur zusammen
- Typ V: Proteine enthalten einen Lipidanker, der die Verankerung in der Membran ermöglicht
- Typ VI: Lipidanker, kovalent und post-translational angeheftet
Glutathion
- Atypisches Tripeptid, besteht aus Glutamat, Cystein und Glycin
- Cystein kann durch SH-Gruppe Disulfidbrücke bilden
- Glutamat ist über eine Isopeptidbindung mit Cystein verbunden
- Funktionen:
- Wichtiges Antioxidans in allen Zellen
- Schutz vor O2-Radikalen in Erythrozyten
- Wichtig bei Entgiftungsreaktionen
