Question 1: Answer all parts You have been asked to design a number of temperature sensitive liposome formulations loaded with the chemotherapeutic doxorubicin. You have been provided the following chemical structures (Tm and PEG molecular weight is given for some chemicals): 2 A) Design a liposomal bilayer, using the given chemical structures, that shows rapid drug release at physiological temperatures. Explain your design choices. (10 marks) B) Design a liposomal bilayer, using the given chemical structures, that shows slow drug release at physiological temperatures. Explain your design choices. (10 marks) C) Design a doxorubicin loaded liposome, using the given chemical structures and any material in your lectures, that will circulate for prolonged periods in the blood stream, be readily engulfed by cancer cells and release drug upon heating to 44 °C. Explain all your design choices and the physiological barriers/mechanisms that must be avoided or minimized to achieve your design goal. (10 marks) Describe each system in as much detail as possible. This should include describing the fundamental science that underpins your design choice, describing the physicochemical properties of the system, describing biological interactions with the system in vivo, and any foreseeable challenges with the system. An example would be to describe the fundamental mechanisms that promote liposome stability both on the bench and in the body. The liposome system can be any size between 50-300 nm and must comprise liposome constituent chemicals as shown above. Question 2: Answer all parts A) Describe in detail the mechanisms by which nanodrugs are uptaken by cells and how these mechanisms alter the fate of the nanodrug within the cell. (15 marks) B) Describe in detail the mechanism by which amphiphilic small molecules self-assemble into larger nanostructures in water.
