To which pH value do I need to adjust my urine sample to have 99% of the compound under its ionized form? If my sample has a pH of 9.3, what is percentage of amphetamine that is under its neutral form? 9 Bioanalytical Sciences 2020-2021 | [email protected] I am interested in extracting amphetamine from urine using SPE, and need the compound to be 99% ionized. To which pH value do I need to adjust my urine sample to have 99% of the compound under its ionized form? The pKa of the amine group of MDMA is 9.9. 13 Bioanalytical Sciences 2020-2021 | [email protected] Q&A INTERMEZZO pH and pKa pH = pKa - 2 I am interested in extracting MDMA from urine using SPE, and need the compound to be 99% ionized. To which pH value do I need to adjust my urine sample to have 99% of the compound under its ionized form? The pKa of the amine group of MDMA is 9.9. At pH lower than 7.9 (9.9 – 2), MDMA is ≥99% ionized. This means that my urine samples has to have a pH lower than 7.9. The pH of urine can be between 3.5 – 8 (usually 4 – 6.5 in non- doped and healthy people). Even though this pH is sufficient to have MDMA ionized, it is better to adjust the pH with a buffer to be sure that all samples have a similar pH (adjust to a value 14 lower than 7.9, the exact value depends on the application).
