Study Notes
Serotonin and Its Role
- Involved in migraine, depression, anxiety, pain, appetite, sleep, and vomiting
- Derived from tryptophan
- Found mainly in enterochromaffin cells in the periphery
Classification of Neurotransmitters
- Neuropeptides: include substance P and endogenous opioids
- Amino acids: include GABA, glutamate, and glycine
- Monoamines: include serotonin, dopamine, and norepinephrine
- Histamine
- Endocannabinoids
- Acetylcholine: found in ganiglia and CNS, has M and N receptors
Serotonin Receptors
- All 5-HT receptors are metabotropic, except 5-HT3 which is ionotropic
- 5-HT1 and 5-HT5: Gi-protein coupled, inhibitory
- 5-HT2 and 5-HT6: Gs-protein coupled, excitatory
- 5-HT3: Ligand-gated Na+ and K+ channel, excitatory
- 5-HT4: Gi-protein coupled, inhibitory
- 5-HT7: Gs-protein coupled, excitatory
Serotonin Agonists and Antagonists
- Agonists: buspirone, triptans
- Antagonists: clozapine, methysergide, ondansetron, cyproheptadine
- Atypical antipsychotics: block dopamine and mainly serotonin receptors, treat both positive and negative symptoms of schizophrenia
Other Neurotransmitters
- GABA: synthesized from glutamate using the enzyme L-glutamic acid decarboxylase and vitamin B6 as a cofactor
