    Study Notes

    Objective

    • Students should extract DNA from bananas
    • Students should learn about genetically modified organisms
    • Students should write a position paper

    Principle

    • DNA is gently separated from other cellular components
    • The Marmur procedure is used
    • Soap solution breaks down fats and proteins
    • Membranes surrounding cells are broken down
    • Salt causes the DNA strands to aggregate
    • Alcohol separates DNA and other components

    Materials

    • 100 mL graduated cylinder
    • 10 mL graduated cylinder
    • 250 mL beaker
    • Test tube (16 x 150)
    • Stirring rod
    • Test tube rack
    • Dropper
    • 15 mL Ethyl Alcohol
    • 150 mL Distilled Water
    • 5 mL Joy dish washing liquid
    • 1 g Sodium Chloride
    • 2 Plastic Cups
    • 2 Plastic Spoons
    • Ice cubes/tubes
    • Blender
    • Small cheese cloth

    Procedure

    • Blend 1 peeled banana with 150 mL water
    • Mix 1 g salt with 10 mL water until dissolved
    • Add 5 mL dish soap to the mixture
    • Add banana mixture to soap solution and stir
    • Filter the mixture
    • Add 10 mL alcohol to a beaker with ice
    • Add filtrate to alcohol using a dropper
    • Gently mix and let stand
    • The glass rod is used to spool out the DNA
