Study Notes
- Total leukocyte count involves counting the different types of leukocytes in one cubic millimeter of circulating blood.
- Blood is diluted 20 times with a white blood cell diluting fluid.
- Diluting fluid causes the lysis of red blood cells, leaving the white blood cells.
Materials required
- Blood sample
- Improved Neubauer chamber with cover slide
- Sahli pipette (0.02 ml)
- Khan tube
- Microscope
- 2% glacial acetic acid as diluting fluid
Diluting fluid preparation
- Diluting fluid is prepared with 2% glacial acetic acid and a drop of methylene blue.
- Methylene blue stains the nuclei of WBC with a blue color.
Normal leukocyte values
- Adult: 4000-11000/c.mm
- Children (1-10): 4,500-13,000/c.mm
- Newborn: 10,000-25000/c.mm
Leukocytosis
- Leukocytosis indicates an increased number of leukocytes.
- Leukocytosis can occur in acute inflammatory processes.
- Leukocytosis can occur in to bacterial, fungal, and parasitic infections.
- Leukemia can cause Leukocytosis.
- Cancer can cause Leukocytosis.
- Exposure to chemicals can result in Leukocytosis.
Leukopenia or Leukocytopenia
- Leukopenia indicates a decreased number of leukocytes.
- Viral infections can cause Leukopenia.
- Chemotherapy can cause Leukopenia.
- Radiotherapy can cause Leukopenia.
- Aplastic anemia can cause Leukopenia.
- Mylofibrosis can cause Leukopenia.
