Media Classification by Consistency

  • Solid media:
    • Advantages: identification of bacteria by colony character, separation of mixed bacteria, and isolation of bacteria as pure culture
    • Agar is commonly used to prepare solid media, which is:
      • Bacteriologically inert
      • Remains solid at 37°C
      • Transparent
  • Liquid media:
    • Used for profuse growth, e.g., blood culture
    • Mixed organisms cannot be separated

Media Classification by Chemical Composition

  • Routine Laboratory Media:
    • Chemically defined media prepared from pure chemical substances
    • Used in research work
  • Synthetic Media:
    • Classified into six types:
      • Basal media
      • Enriched media
      • Selective media
      • Indicator media
      • Transport media
      • Storage media

Synthetic Media Types

Basal Media

  • Used for growth of bacteria that do not need enrichment
  • Examples: Nutrient broth, nutrient agar, and peptone water
  • Suitable for growth of Staphylococcus and Enterobacteriaceae

Enriched Media

  • Enriched by adding blood, serum, or egg
  • Examples: Blood agar and Lowenstein-Jensen media
  • Suitable for growth of Streptococci

Selective Media

  • Favour growth of a particular bacterium by inhibiting undesired bacteria
  • Examples: MacConkey agar, Lowenstein-Jensen media, and tellurite media
  • Antibiotic may be added to inhibit growth of undesired bacteria

Indicator Media

  • Contain an indicator that changes in response to a particular organism
  • Examples: Blood agar and MacConkey agar
  • Indicator media are also known as differential media
