- The liver is the primary organ for drug metabolism, with other tissues—lungs, intestines, kidneys, and skin—also involved in drug biotransformation. Metabolism may occur in other tissues to varying degrees depending on drug properties and administration route.
- The liver is organized into lobules, containing parenchymal cells within a network of blood and lymphatic vessels.
- The liver has large right and left lobes that merge centrally.
- Blood supply sources include the hepatic artery (~25% of supply) and the portal vein (~75% of supply).
- Blood from these vessels mixes in the sinusoids before exiting via the hepatic vein to the vena cava.
Bile Production and Secretion
- The liver secretes bile acids, which flow through channels into the common bile duct.
- The common bile duct drains bile and biliary excretion products from both lobes into the gallbladder.
Drug Metabolism in the Liver
