Histopathologic Techniques
- Cryostat is an apparatus used in fresh tissue microtomy (frozen sections) and is left on even when not in use.
- Optimum working temperature of Cryostat is -18-20°C.
- Temperature situations affect the quality of tissue cutting:
- Tissue < Knife: Compression displaced structures
- Tissue > Knife: Melts tissue, sticky & distorted mass
- Tissue >> Knife: Resistant to cutting, brittle, fragments

- Histology is the microscopic study of normal tissues of the body.
- Histopathology is the branch of pathology dealing with diseased tissues.
Types of Samples
