Describe the various methods involved in tissue processing. 01 | Introduction to Human Histology Histology ○ Histo = tissue + logia = science ○ Microscopic anatomy ○ Scientific study of microscopic structures of tissues and organs of the body Significance of Histology to MedTech Profession In the Philippines RMTs assist pathologists in the laboratory Preparing and processing tissue sample Significance of Histology to MedTech Profession In the United States Can be histotechnician HT(ASCP) or histotechnologist HTL(ASCP) ○ Histotechnician - Prepares and processes tissue samples for routine histologic techniques (fixation, processing, embedding, microtomy, staining) ○ Histotechnologist - Performs both routine and complex histologic techniques (enzyme histochemistry, immunohistochemistry, in-situ hybridization, immunofluorescence) Significance of Histology to MedTech Profession In the United States Can be histotechnician HT(ASCP) or histotechnologist HTL(ASCP) ○ Requirements Education: obtaining associate or bachelor’s degree or completing an accredited histologic technician program Certification: certification form ASCP REVIEW Levels of Organization in the Human Body O_______ O____ ______ O____ T_____ C___ C_______ 02 | Tissue Processing 1. FIXATION Small pieces of tissue are placed in solutions of chemical that cross-link proteins and inactivate degradative enzymes, which preserve cell and tissue structure. Aim: preserve the tissue nearest to its living state Commonly used fixative: formalin 2. DEHYDRATION The tissue is transferred through a series of increasingly concentrated alcohol solutions, ending in 100% alcohol which removes all water. Aim: remove water molecule of the tissue Commonly used dehydrating agent: ethanol 3. CLEARING Dehydrating agent is removed and replaced by an organic solvent to facilitate the transition of dehydration and infiltration stage. Aim: removal of dehydrating agent, make tissue clear and improve microscopic exam Commonly used clearing agent: Xylene 4. INFILTRATION The tissue is then places in melted medium until it becomes completely infiltrated. AIM: to provide support to the tissue, permit thin sectioning Commonly used medium: Paraffin wax 5. EMBEDDING The infiltrated tissue is placed in a small mold with melted media and allowed to harden. AIM: to provide support to the tissue, permit thin sectioning Commonly used medium: Paraffin wax 6. TRIMMING The resulting paraffin block is trimmed to expose the tissue for sectioning on a microtome. 7. SECTIONING Uses microtome that cuts tissue specimen into thin paraffin ribbon that can be placed on a slide for microscopy. 7. SECTIONING 8. WATER BATH The tissue ribbon is floated on a water bath to flatten it, then fished onto a microscope slide coated with Meyer’s egg albumin as an adhesive. 9. STAINING The tissue will subsequently be stained to visualize different tissue components. Commonly used stain: Hematoxylin and Eosin 10. MOUNTING A mounting agent will then be added to the specimen and a thin glass coverslip placed over the specimen to secure it and prevent damage.
