Protein Delivery Learning objectives: 1. Describe structure of proteins 2. Describe stability issues of proteins 3. Identify the differences between small molecule drugs and proteins 4. Describe different types of proteins 5. Explain how formulations are developed for proteins Proteins  Proteins are polypeptides composed of covalently linked amino acids.  Polypeptides with >40 amino acids are called proteins.  Nearly 380 FDA approved proteins, and a total 60 peptide- based drugs are already in the market. (PLoS One. 2017; 12(7): e0181748)  Amino acids are zwitter ionic at pH 7 because of ionization of both the acid and the amine groups. Such a molecule containing both positive and negative charge is called amphipathic. Building blocks of proteins Hydrophilic Hydrophobic Can form 130 variants of peptide bond formation. No clear definition but >40 AA are proteins;
