GUIDELINES IN THE LABORATORY BTC-104L CHEMICAL BIOLOGY II (Analytical Methods for Biology – Laboratory) Students enrolled in BTC-104L CHEMICAL BIOLOGY II (Analytical Methods for Biology – Laboratory) courses are required to observe the guidelines given below: A. GENERAL REQUIREMENTS 1. Students are required to have a laboratory gown, laboratory manual and laboratory notebook in every laboratory period. 2. Each student is also required to write the pre-lab data in their respective laboratory notebook before coming to class. The format for the pre-lab data is given in the succeeding section. Failure to comply with the above requirements will mean a grade of zero for the laboratory notebook, laboratory data, laboratory performance, and laboratory report sheet. B. LABORATORY HOUSEKEEPING & SAFETY RULES 1. Always wear a laboratory gown while in the laboratory. 2. Students are strictly prohibited from eating, drinking, loitering and smoking inside the laboratory. 3. During laboratory period, students are strictly prohibited from entertaining students from other class or staying in the lobby not unless students are borrowing/returning the apparatus. 4. Students are allowed to perform only the scheduled experiment. Unauthorized experiment should not be performed. 5. Students are encouraged to be familiar with the safety features of the laboratory such as the fire extinguisher, medicine cabinet, and nearest fire exit. 6. Make sure the complete list is prepared upon requisition of glassware and other materials. Check each item for cracks and other defects. Defective glassware and materials must be reported immediately. 7. Always use reagents, water and gas economically. 8. Do not touch, taste or smell chemicals unless otherwise directed to do so. Should an odor test be necessary, do not inhale the vapour directly from the test tube but instead, waft a bit of vapor towards your nose. 9. Do not contaminate chemicals. Use the dropper or spatula designated for each reagent bottle. 10. Corrosive acids and strongly caustic substances should be carefully handled. Spilled acids on the skin are neutralized with sodium bicarbonate; while strong bases, with dilute acetic acid and then washed off immediately with plenty of water. 11. When preparing dilute acid solutions, always add acid to water, never water to acid. 12. When heating, always use a non-luminous flame that is not more than two inches high. Always turn-off the gas cock when the Bunsen burner is not in use. 13. Always carefully set aside heated objects to cool. 14. Protect your hands with pot holder when a glass rod, glass tubing, or a thermometer is to be inserted or removed from stoppers. 15. Report all accidents, even minor ones, to the instructor. 16. Insoluble refuse should be thrown in the waste can, not into the sink. 17. Waste acids should be poured into the disposal jar not into the sink. 18. Use the fume hood whenever toxic and irritating gases are generated during the experiment. 19. Record observations and data immediately in your laboratory manual 20. At least 20 minutes before the end of the laboratory period, all glass wares must be cleaned and dried properly. Likewise, clean the working area to make it ready for the next users. 21. Please put all the chairs on top of the table and check that all lights, ceiling fans, water and gas cocks near your working area are turned off before leaving the laboratory. Lantajo, Ph.D. GENERAL DIRECTIONS: Read carefully the entire experiment before coming to the laboratory, and familiarize yourself with the theory involved as well as the experimental part. YOU ARE EXPECTED TO KNOW WHAT YOU ARE DOING AT THE TIME YOU ARE DOING IT. Follow the laboratory directions carefully. If you are in doubt as to the meaning of any terms or directions, consult your instructor. Do not follow directions mechanically but know the reasons for each step. In other words, know why as well as how. Develop your powers of observation and reasoning. Always bring your textbook to the laboratory. You will need it for reference purposes. Record each observation at the time you perform the experiment. In order to complete the laboratory work assigned for the course, it will be necessary to save time wherever possible. Plan your work ahead each laboratory activity. Certain experiments must be completed or carried to a certain stage during the same laboratory period in which the work is begun. Such experiments should be started at the beginning of the period. REACTIONS OFTEN TAKE PLACE WITH EXPLOSIVE VIOLENCE. Therefore always be cautious in your manipulation. Mix reagents only as directed. To do otherwise may result in a violent reaction or explosion. Exactness, neatness and cleanliness are essential to good work and good results. Each student should provide himself with laboratory gown. While working in the laboratory, keep your laboratory guide in such position that no chemical will be spilled on it. Always work with the group unless otherwise directed by the instructor. Take proper care in the use of balances, reagents and other laboratory equipment.
