Mechanism and Physiologic Actions
- Synthesized in the hypothalamus and released from the posterior pituitary gland.
- Regulates water retention in kidneys by increasing water reabsorption.
- Acts through V1 and V2 receptors; V2 receptors mediate water conservation, while V1 receptors cause vasoconstriction.
- Increases permeability of kidney collecting ducts to water and can cause vasoconstriction, especially in cardiovascular smooth muscle.
Clinical Applications
- Used for various conditions including:
- Diabetes Insipidus
- Gastrointestinal hemorrhage
- Esophageal varices with bleeding
- Cardiac arrest conditions such as asystole, ventricular fibrillation, and ventricular tachycardia.
- Administered for abdominal distension before radiological procedures.

- Metabolized quickly with a plasma half-life of about 10 to 20 minutes.
- Up to 5% of the administered dose can be excreted unchanged in urine after 4 hours.
Preparation and Dosage Forms
- Available in 1 mL vials with 20 pressor units per mL for intramuscular, subcutaneous, and intravenous administration.
