Tissue Impregnation/Embedding

  • Infiltration/impregnation involves replacing the clearing medium with a support medium that fills all tissue cavities.

Types of Tissue Impregnation/Embedding

  • Paraffin wax: simplest, most common, and best method
  • Celloidin (collodion): used for bones, teeth, large brain sections (wet), and whole eye sections (dry)
  • Gelatin: another type of impregnation/embedding
  • Plastic: also used for impregnation/embedding

Paraffin Wax Impregnation

  • Melting point: 54°C-58°C at 20°C-24°C
  • Advantages: ease in cutting, permanent paraffin blocks, and good staining results
  • Disadvantages: not recommended for fatty tissues, overheated paraffin leads to brittle specimens, and inadequate impregnation retains clearing agent

Substitutes for Paraffin Wax
