DNA Replication
Initiation
- DNA replication begins at specific regions called origin of replication
- Unwinding proteins (helicases) unwind the double helix, creating a replication fork
- An RNA molecule (primer) binds to the template strand at the replication fork
- Helicase, primase, and DNA polymerase bind to the replication fork
Unwinding
- Helicase unwinds the double helix, creating a replication fork
- Topoisomerase relaxes the tension in the DNA molecule, preventing it from getting tangled
- Single-strand binding proteins stabilize the unwound strands, preventing them from rewinding
Elongation
- DNA polymerase adds nucleotides to the growing strand, matching the base pairing rules
- The leading strand is synthesized continuously in the 5' to 3' direction
- The lagging strand is synthesized in short, discontinuous segments (Okazaki fragments) in the 5' to 3' direction
- RNAse removes the RNA primers, and DNA polymerase fills the gaps
Proofreading
- DNA polymerase has a built-in proofreading activity, checking for errors during synthesis
- Exonuclease activity corrects mismatched nucleotides, ensuring the correct sequence
- Mismatch repair corrects errors after replication, ensuring the integrity of the genetic information
Termination
- The replication process is complete when the replication fork converges
- RNAse removes the RNA primers, and DNA ligase seals the gaps, forming a continuous double-stranded DNA molecule
- Topoisomerase relaxes the tension in the newly synthesized DNA molecule, ensuring its stability
