Study Notes
Medical Microbiology - Microbial Growth and Nutrition
- Medical microbiology is the study of microorganisms that cause disease in humans.
- Microorganisms require specific physical, gaseous, and chemical conditions for optimal growth.
Microbial Growth Curve
- Bacterial growth progresses through distinct phases:
- Lag phase: initial period of adjustment to new environment, no increase in cell number
- Log (exponential) phase: rapid cell division; cells multiply at constant rate; measured on log scale, exponential growth
- Stationary phase: rate of cell division equals rate of cell death, no net change in cell number
- Death phase: cells die off at a constant rate; logarithmic decline
Bacterial Growth Requirements - Physical
-
Temperature:
- Minimum growth temperature: the lowest temperature at which bacteria can grow.
- Optimum growth temperature: the temperature at which bacteria grow most rapidly.
- Maximum growth temperature: the highest temperature at which bacteria can grow.
- The temp range is the range between the minimum and maximum temperatures. Growth does not occur outside of this range
-
Classification of Bacteria based on Temperature:
- Psychrophiles: cold-loving bacteria, optimal growth below 20 °C.
- Psychrotrophs: cold-tolerant bacteria, grow best between 0 and 30 °C and tolerate cold
- Mesophiles: moderate-temperature loving bacteria, optimal growth between 10-48 °C; includes most human pathogens (body temperature)
- Thermophiles: heat-loving bacteria, optimal growth between 40-72 °C
- Hyperthermophiles: extreme heat-loving bacteria, optimal growth between 65-110 °C
-
pH:
- Majority of bacteria are neutrophiles and grow best between pH 6.8 - 7.4
- Acidophiles: grow best at low pH (less than 6.8)
- Alkalophiles: grow best at high pH (above 7.4)
-
Osmotic Pressure:
- Osmotolerant/halotolerant/barotolerant bacteria can grow in isotonic or hypotonic environments and can tolerate high osmotic pressure (but do not require it).
- Obligate or extreme halophiles/osmophiles require relatively high salt concentrations for growth.
-
Water Activity (aw): aw is a measure of water availability; the ratio of the vapor pressure of a solution to the vapor pressure of pure water (always 1.00 for pure water ).
Bacterial Growth requirements - Gaseous
-
Oxygen:
- Obligate aerobes require oxygen to grow
- Facultative anaerobes can grow in the presence or absence of oxygen.
- Aerotolerant anaerobes tolerate oxygen but do not require it for growth
- Microaerophiles need very low levels of oxygen to grow.
- Obligate anaerobes cannot grow in the presence of oxygen.
-
Carbon dioxide (CO₂):
- Most bacteria can grow with low levels of CO2.
- Capnophiles require high CO2 levels to grow.
Bacterial Growth requirements - Chemical
- Microbes have different nutritional needs which affect bacterial growth
