- Size enlargement deals with making portions of matter larger than component particles
- Methods include granulation, flocculation, extrusion, compression, and coating
- Objectives of size enlargement include: -preventing air trapping in tableting and varying powder densities -increasing flowability in hoppers and dies -increasing bulk density for easier storage and transportation -preventing dust losses during drying, filling, or shipping -reducing dust nuisance and hazard -reducing surface area for hygroscopic or reactive substances to minimize water absorption and enhance stability
Granulation Methods
- Granulation methods are divided into two types: -Wet methods (using liquid) -Dry methods (no liquid)
A. Wet Granulation
- Involves massing powder mix using a non-toxic and volatile solvent
- Methods include: -Shear granulation (using a planetary mixer like Hobart Mixer) -Granulation by fusion (using suitable wax binders) -Spray granulation (by introducing granulating liquid and drying) -Granulation by extrusion (similar to mincing meat) -Granulation by spheronisation (using spheronising or pelletising equipment like Freund Granulator)
B. Dry Granulation
- Methods include: -Slugging by Tablet Presses (compressing powder into dies of a tablet press) -Roller Compaction (feeding the powder between rollers to form compressed sheets)
- Used when active ingredients are sensitive to moisture, heat, or both
