    Study Notes

    • The purpose of this procedure is to provide instruction in the fixation and processing of tissue specimens for Histology diagnosis.
    • Any tissue removed from the body and submitted to Histology for diagnosis is eligible for use in this procedure.
    • Technicians will conduct and maintain documentation of preventive maintenance and quality control on all instrumentation and equipment used to perform this procedure.
    • Technicians will perform validations for new tissue processors (AP-HIS.708). Revalidations are performed if tissue processors are moved or after repairs have been completed.
    • A panel of tissue should be processed and validated by a qualified pathologist.
    • All tissue samples that are submitted for histology must be immersed in 10% NBF and processed on the 8 or 12 hour program cycle.
    • Bloody, poorly fixed and fatty specimens will be processed on the 12 hour program cycle.
    • Tissue samples that have been placed in Bouin’s fixative, Potassasium Hydroxide or Rapid Decalcifier (RDO) must be rinsed before processing.
    • Loading/unloading specimens onto processors: 1. Open the retort lid by turning the handle counter-clockwise to the unlock position and open the retort lid. Load the baskets with the cassettes inside (text facing up) and covered with a basket lid into the retort. Close the retort lid and turn the handle clockwise to the lock position.
    1. From the main screen, log on by using the number keypad. Enter the code 100000, and then press ENTER. Press MENU. The Process Menu will appear.
    2. Using the Process Menu, press TISSUE PROCESSING. Select the desired program from the from the Program List. Refer to Procedure Notes for tissue processing programs.
    3. To modify the predicted end time, press EDIT PROGRAM, press EDIT, press EDIT PROGRAM, select BAMC and press SAVE. NOTE: Every program will be on a 24 hour clock (Military Time).
    4. Press PREDICTED ENDTIME, then input the predicted end time and or date, then press SAVE. Press SAVE again, then press START, and then press START again.
    5. When tissue processing is complete, a notification window will automatically be displayed. Press EXIT, and Press REMOVE BASKETS. Press YES to drain the retort. (Remove all baskets and cassettes).
    6. The Clean Retort tab will automatically be displayed. To clean retort, press START, and then press YES.
    • The AP-HIS-205 Fixation and Processing (HIS) software is approved and current.
    • The program is available on all tissue processors.
    • Normally a five-hour process can be adjusted according to tissue size, thickness, and prior fixation time.
    • If embedded, the block must be melted down and the specimen placed in a cassette labeled with the correct accession number.
    • The gross room technician will process the block with the rest of the blocks that are placed on the 12-hour program for that day.
    • Blocks that are reprocessed in CoPathPlus.F must be scanned as such. If there are any issues with the tissue processor, please contact the tissue-tek VIP 6 Vaccum Infiltration Processor Operating Manual lead technician or supervisor.
    • When opening a new lot number of Buffered Formalin, Certificate of Analysis must be printed if it hasn't been already been done and record the lot and your initials on the pH formalin Certificate of Analysis Log (AP-HIS.A207).

    • Then click on Resources at the top of the page.
    • Click on Search Documents.
    • Click on Epredia Formalin COAs.
    • Click on the pdf that contains the lot number listed next to it (11111000.pdf).
    • Make sure the ph is in range (6.9-7.1) and print the document.
    • File the COA in the notebook.
    • Fill out the pH Formalin Certificate of Analysis Log (AP-HIS.A207).
