    Study Notes

    Introduction to Histology

    • Histology is the study of the microscopic anatomy of cells and tissues.
    • It involves the examination of tissues using microscopy.
    • The goal is to understand the structure and organization of cells and tissues to understand their function.
    • Histological techniques are used to preserve, process, and stain tissue samples for viewing under a microscope.

    Tissue Preparation Techniques

    • Fixation: Preserves tissue structure and prevents decay. Common fixatives include formaldehyde and glutaraldehyde.
    • Dehydration: Removes water from the tissue, preparing it for embedding. Alcohol is a common dehydrating agent.
    • Clearing: Replaces alcohol with a clearing agent, like xylene, which is miscible with paraffin.
    • Embedding: The cleared tissue is embedded in a supporting medium, typically paraffin wax.
    • Sectioning: The embedded tissue block is sliced into thin sections using a microtome.
    • Staining: Sections are then stained to enhance contrast and visibility of different cellular components. Common stains include H&E (hematoxylin and eosin).
    • Mounting: The stained sections are mounted onto slides for viewing under a microscope.

    Common Tissue Types & Characteristics

    • Epithelial Tissue: Covers surfaces, lines cavities, and forms glands.
      • Characterized by tightly packed cells, minimal extracellular matrix, and usually a basement membrane.
      • Functions include protection, secretion, absorption, and excretion.
      • Different types include squamous, cuboidal, columnar, pseudostratified, transitional.
    • Connective Tissue: Supports, connects, and separates different tissues and organs.
      • Characterized by a large amount of extracellular matrix, which provides support and strength.
      • Different types include loose connective tissue, dense connective tissue, cartilage, bone, blood.
      • Functions include support, protection, transport, and storage.
    • Muscle Tissue: Enables movement.
      • Characterized by contractile proteins (actin and myosin).
      • Three types of muscle tissue: skeletal, smooth, and cardiac.
      • Functions include movement, support, and pump blood.
    • Nervous Tissue: Detects, processes, and transmits information.
      • Characterized by neurons and neuroglia.
      • Functions include communication, control, and coordination.

    Staining Techniques & Applications

    • Hematoxylin and Eosin (H&E): A common stain used for general histology.
      • Hematoxylin stains the nuclei of the cell dark blue/purple.
      • Eosin stains the cytoplasm and other extracellular components a pinkish color.
    • Special stains: Used to highlight specific components or structures within tissues.
      • Examples include periodic acid-Schiff (PAS) stain for carbohydrates, Van Gieson stain for collagen, and silver stains for nerve fibers.
    • Immunohistochemistry: Uses antibodies to detect specific proteins within the tissue, allowing identification of particular cells or to study the presence and distribution of molecules.

    Microscopy Techniques

    • Light Microscopy: Allows visualization of stained tissue sections.
    • Electron Microscopy (TEM & SEM): Provides high-resolution images of cellular structures.
      • Transmission electron microscopy (TEM): Shows internal structures at a high level of detail.
      • Scanning electron microscopy (SEM): Shows surface structures in detail.

    Relevance to other disciplines

    • Histology provides essential insight into the structure and function of biological systems.
    • This understanding is critical to several fields like pathology, physiology, and cell biology.
    • Histological examinations are essential in the diagnosis of disease.
    • The ability to identify tissue types and abnormalities can assist physicians in treatment and management of different conditions.
