Normal SPE (Serum Protein Electrophoresis)
- Electrophoresis separation of serum protein mixture on cellulose acetate shows five bands, in order of highest to lowest migration toward the anode:
Bands and Reference Ranges
- Albumin: 35-52 g/L
- α1-globulin: 2-4 g/L
- α2-globulin: 5-9 g/L
- β-globulin: 6-11 g/L
- γ-globulin: 7-17 g/L
Composition of Each Band (Except Albumin)
- α1-globulin: contains α1-antitrypsin, α1-acid glycoprotein, α1-fetoprotein, HDL, and others
- α2-globulin: contains α2-macroglobulin, haptoglobin, and others
- β-globulin: contains transferrin, β2-microglobulin, plasminogen, LDL, and others
- γ-globulin: contains mainly immunoglobulins (IgG, IgA, IgM, and to a lesser extent IgD and IgE)
Important Note
- If plasma is used instead of serum, an additional fibrinogen band appears between β and γ-globulin bands, which can be confused with the M-band, so serum should be used in protein electrophoresis.
