    Study Notes

    Cells of the Immune System

    • Leukocytes are the main cells of the immune system
      • Lymphocytes are part of the lymphoid lineage
        • Several types of lymphocytes exist
      • Myeloid lineage contains:
        • Dendritic cells
        • Monocytes/Macrophages
        • Granulocytes

    Haematopoiesis

    • All leukocytes are generated in primary lymphoid tissue by the process of haematopoiesis
    • Lymphoid and myeloid cells follow different differentiation pathways
    • Haematopoietic stem cell (HSC) is the origin of both lymphoid and myeloid cells

    Granulocytes

    • Front line of attack during an immune response, part of the innate immune system
    • Identified by characteristic staining patterns of “granules”
    • Released in contact with pathogens
    • Contain proteins with distinct functions: killing, regulation of other cells, tissue remodeling
    • All have multilobed nuclei

    Neutrophils

    • One of the main effector cells in the innate immune system
    • Constitutes 50-70% of white blood cells
    • Released from bone marrow, circulate for 7-10 hours, enter tissues, live only a few days
    • Numbers and recruitment increase during infections, called "leukocytosis"
    • Identified by phagocytosis of microorganisms
    • Main cellular component of pus

    Neutrophil Characteristics

    • Named based on staining qualities of granules
    • Multilobed nucleus is called a polymorphonuclear leukocyte (PMN)
    • Neutrophilic granules stain lightly blue to pink
    • Spend 7-10 hours in the blood before migrating into tissues
    • First responders, motile and phagocytic
    • "Leukocytosis" indicates infection
    • Release extracellular "traps"

    Basophils

    • Basophils are part of the granulocyte lineage
