- PCR is based on using the ability of DNA polymerase to synthesize new strand of DNA complementary to the offered template strand. - It is widely used in molecular biology to make several copies of a specific DNA segment. Free DNA Preparation 1- PCR primers are short chain of single-stranded DNA, usually around 20 nucleotides in length. Two primers are used in each PCR reaction. 2- Deoxynucleoside triphosphates (dNTPs) consist of four basic nucleotides—dATP, dCTP, dGTP, and dTTP—as building blocks of new DNA strands. 3- The function of Taq DNA polymerase in PCR reaction is to amplify the DNA for the production of multiple copies of DNA.
