Microscope ▪Importance ▪Types ▪Parts IMPORTANCE: ⚫The microscope is a very important tool in studying living things. ⚫Scientists are able to study cells and the wide array of microscopic organisms. Types of Microscopes 1. Compound light microscope - commonly use in classroom work. This uses light to magnify the specimen. Common magnifications: 40x, 100x, 400x Types of Microscopes 2. Stereomicroscope (dissecting microscope) - used for dissecting small specimen. It has a lower magnification than the LCM. 3. Phase contrast microscope - utilizes the refraction of light in contrast to the LCM which uses direct light. MORE POWERFUL (M) 1.Transmission Electron Microscope (TEM) - uses electrons instead of light as a light source. Magnification 100,000 times. 2. Scanning Electron microscope (SEM) - uses a focused beam of high-energy electrons to generate a variety of signals at the surface of solid specimen. Magnification more than 500,000 times. Eyepiece Body Tube Revolving Nosepiece Arm Objective Lens Stage Stage Clips Coarse adjustment knob Diaphragm Fine adjustment knob Light Base MECHANICAL PARTS OF A MICROSCOPE 1. Base- supports the entire microscope. 2. Arm- the part which you hold to move or carry the (M). 3. Body tube- connects the upper part of (M) to its lower parts. MECHANICAL PARTS OF A MICROSCOPE 4. Revolving nosepiece- used to shift the objective lens. 5. Coarse adjustment knob- used for raising or lowering the body tube when one is focusing, using the (LPO). 6. Fine adjustment knob- used for fine focu- sing when using the (HPO). MECHANICAL PARTS OF A MICROSCOPE 7. Stage- where the glass slide is placed. 8. Stage clip- used to hold the slide in place. OPTICAL PARTS OF A MICROSCOPE 1.Mirror/Light- directs the light up through the object. 2.Diaphragm- regulates the amount of light reflected by the mirror. 3. Condenser- concentrates the light on the object. 4. Low power objective (LPO)- magnifies the object 10 to 15 times and gives the object being OPTICAL PARTS OF A MICROSCOPE 5. High power objective (HPO)- magnifies the object 40-45 times. 6. Oil immersion objective- magnifies the object 100 times and makes use of a special kind of oil, the cedar oil. 7. Ocular or Eyepiece- part where one looks through to view the specimen being focused. Practice Labeling the Parts
