Trazadone is an SARIS (serotonin antagonist reuptake inhibitor) that works by blocking 5-HT & 5-HT 2C receptors allowing more 5-HT to interact at postsynaptic 5-HT sites. SARIS is affected by Serotonin blockade & reuptake inhibition at higher doses. Trazadone. Trazadone also blocks histamine and A-adrenergic receptors are the most common SARIs Trazadone is an adjunctive treatment for clients with MDD who report difficulty falling or staying asleep Off label use for Trazadone is insomnia & anxiety, therefore should be taken at bedtime Sedation, drowsiness, blurred vision, constipation, & dry mouth are common side effects of Trazadone Warn patients of the risk for priapism (a medical emergency) when taking trazadone Neurotransmitters that affect tricyclic antidepressants (TCAs) include both SRI & NRI works by blocking a-adrenergic, histamine-1, & muscarinic cholinergic receptors TCAs are not used first line because of the high incidence of adverse effects & risk for potential overdose & death due to overdose constipation, urinary retention, weight gain, blurred vision, dry mouth, sedation, orthostatic hypotension are common side effects of TCA When other central nervous system depressants are prescribed avoid TCAs
