Tooth Development - Initiation
- Tooth development is complex and involves epithelial-mesenchymal interactions, morphogenesis, and mineralization.
- Humans develop 20 deciduous and 32 permanent teeth.
- Tooth development begins with the interaction of oral epithelium (ectoderm) and neural crest cells derived mesenchyme (mesoderm)
- Neural crest cells migrate from the lateral parts of the neural tube during the 3rd week of embryonic development and enter the underlying mesenchyme, now termed ectomesenchyme.
- Vitamins, minerals, and hormones influence tooth development:
- Vitamin A is important for epithelial growth.
- Vitamin C is important for connective tissue development.
- Vitamin D is essential for calcification.
Tooth Development - Phases
- Tooth development is divided into three overlapping phases:
- Initiation: Sites of future teeth are established with an invagination of oral epithelium called the dental lamina.
- Morphogenesis: The shape of teeth is determined by cell proliferation and movement.
- Histogenesis: Differentiation of cells begun during morphogenesis leads to the formation of fully formed dental tissues, both mineralized (enamel, dentin, and cementum) and unmineralized (pulp and periodontal ligament).
Primary Epithelial Band
- After 37 days of development, a continuous band of thickened epithelium forms around the mouth in the presumptive upper and lower jaws.
- This horseshoe shaped band is known as the primary epithelial band.
- The primary epithelial band gives rise to two subdivisions:
- Dental lamina: Forms first and marks the future positions of teeth.
- Vestibular lamina: Forms shortly after and is positioned just in front of the dental lamina.
Dental Lamina
- The dental lamina is a band of epithelium that invades the underlying ectomesenchyme along the horseshoe-shaped future dental arches.
- Deciduous teeth develop directly from the dental lamina at the eighth week of fetal life.
- Permanent molars develop from a distal extension of the dental lamina.
- Along the dental lamina, 20 areas of enlargement appear, representing the forming buds of the 20 primary teeth.
- The dental lamina continues to develop the 32 permanent tooth buds.
- The lingual extension of the dental lamina is called the successional lamina.
- The successional lamina is responsible for the development of permanent incisors, canines, and premolars.
- The successional lamina is active from the fifth month in utero (for the permanent central incisor) to ten months of age (second premolar).
Vestibular Lamina
- The vestibule forms as a result of the proliferation of the vestibular lamina into the ectomesenchyme.
- Cells of the vestibular lamina rapidly enlarge and then degenerate to form a cleft that becomes the vestibule between the cheek and the tooth-bearing area.
Stages of Tooth Development

- Bud stage: The initial stage of tooth development.

Bud Stage
- The bud stage represents the initiation of tooth development.
- The basement membrane separates the epithelium of the dental lamina from the ectomesenchyme.
- Ten small, round or ovoid swellings develop superficial to the basement membrane called tooth buds.
- Tooth buds are the precursors of enamel organs.
- The epithelium of the tooth bud forms the enamel.
