Study Notes
Stanbio Glucose LiquiColor® (Oxidase) Procedure No. 1070
- Intended use is for quantitative determination of glucose in serum, plasma or CSF
- Reagent contains sodium azide
- Reagent is for in vitro diagnostics, normal lab handling precautions should be taken
- Reagent is ready to use
- The reagent is stable up to the end of the expiration date, if stored correctly at 2-8 °C
- Do not freeze reagent!
- Measurement is not influenced by reagent color changes as long as the absorbance is less than 0.80 at 500 nm in a 1 cm lightpath
- Discard reagent if it contains particulate matter
- Standard is stable up to the end of its expiration date when stored at 2-8 °C
Interfering Substances
- High levels of ascorbic acid can produce falsely low glucose values
Automated Procedure
- Consult your instrument manual for programming instructions
- Specific programming applications for most automated analyzers are available from Stanbio's Technical Service Department
Manual Procedure
- Warm reagent to ambient temperature, zero spectrophotometer at 500 nm with distilled water
- Add 1.0 mL reagent to cuvettes/test tubes and warm to 37 °C for 5 minutes
- Add 10 μL (0.010 mL) of sample to respective cuvette/test tube, mix gently and return to 37 °C incubation
- After 5 minutes of incubation, read and record the absorbance of all samples
- Linearity to 500 mg/dL can be achieved by increasing the reagent volume if desired
Summary and Principle
- Accurate estimation of glucose is important for diagnosis and management of hyperglycemia and hypoglycemia
- Hyperglycemia may occur due to diabetes mellitus, IV glucose fluids, severe stress, or cerebrovascular accidents
- Hypoglycemia may be the result of an insulinoma, insulin administration, inborn error of carbohydrate metabolism, or fasting
- Glucose oxidase methodology was introduced by Keilin and Hartree in 1948
- Keston later reported the use of the combined methodology
