Study Notes
Liquid Dosage Forms
- Liquid dosage forms are formulations containing one or more APIs and excipients in a liquid state.
- Liquid dosage forms can be solutions, suspensions, or emulsions.
- Liquid dosage forms are used for both internal and external use.
Advantages and Disadvantages of Liquid Dosage Forms
- Advantages: easy to administer, accurate dosing, fast acting, versatile.
- Disadvantages: taste issues, limited stability, difficulty administering to certain populations, inconvenient for travel and transportation.
Pharmaceutical Solutions
- Solutions are homogenous, molecular mixtures of two or more components.
- Pharmaceutical solutions consist of a solute (drug and excipients) dissolved in a liquid solvent system.
- Solutions are administered by various routes, including oral, otic, and parenteral.
- Solutions are classified based on the formulation or traditionally by the solvent system, such as syrups, elixirs, spirits, and tinctures.
Traditional Terms for Different Pharmaceutical Solutions
- Aromatic Waters: saturated aqueous solutions of volatile oils or aromatic substances.
- Elixirs: solutions containing alcohol as a cosolvent.
- Spirits: alcoholic or hydro-alcoholic solutions of volatile substances, used as flavoring agents or for medicinal purposes.
- Tinctures: alcoholic or hydro-alcoholic solutions prepared from vegetable materials or chemical substances.
- Syrups: oral aqueous solutions with high concentrations of sucrose or other sugars.
Advantages of Pharmaceutical Solutions
- Solutions provide rapid drug action.
- Solutions allow for accurate dosing through specific volume measurements.
- Oral solutions are easily swallowed, beneficial for children and older individuals.
- Solutions are easier to manufacture than other dosage forms.
Disadvantages of Solutions
- Many drugs are unstable in solution.
- Many drugs are poorly soluble in water.
- Solutions are bulky and less convenient for patients to carry than solid dosage forms.
- Solutions are more expensive to transport.
- Packaging requires higher quality materials.
Excipients Used in Pharmaceutical Solutions
- Excipients enhance product stability, bioavailability, appearance, aid in product manufacture, and identification.
- Excipients should be stable, cost-effective, inert, nonirritant, and compatible with the formulation and route of administration.
Excipients Used in Pharmaceutical Solutions: Flavouring Agents
- Flavouring agents mask the taste of drugs.
- Sweeteners like sucrose, sorbitol, and mannitol are used to improve palatability.
- Synthetic and natural flavourings like vanilla, cinnamon, and menthol are used in oral and nasal solutions.
- Flavour choice should consider patient population and cultural preferences.
Excipients Used in Pharmaceutical Solutions: Colouring Agents
- Colouring agents enhance patient acceptability.
- Colour should correlate with the flavour (e.g., green with mint, red with cherry).
- Labels should have strong adhesive strength and be resistant to water and UV light.
Labelling
- The FDA requires a "Drug Facts" table on all pharmaceutical product labels.
- The table includes:
- Active ingredients and their amounts per dosage.
- Uses of the medicine.
- Directions for taking the medicine.
- Other information, including storage instructions.
- Inactive ingredients.
- Warnings, including:
- "Do Not Use" statements.
- "Ask a Doctor" or "Ask a Doctor or Pharmacist" statements.
- "When Using This Product" statements for side effects and drug interactions.
- "Stop and Ask a Doctor If" and "Stop Using If" statements for serious side effects.
- "Keep Out of Reach of Children" statement.
