  • Staph. aureus is Gram-positive and exhibits beta-hemolysis on blood agar.
  • Staph. epidermidis is Gram-positive and shows gamma-hemolysis on blood agar.
  • Staph. saprophyticus is Gram-positive and presents gamma-hemolysis on blood agar.
  • Micro. luteus is Gram-positive.
  • Strep. pyogenes is Gram-positive and displays beta-hemolysis on blood agar.
  • Strep. agalactiae is Gram-positive and has beta-hemolysis on blood agar.
  • E. faecalis is Gram-positive and exhibits gamma-hemolysis on blood agar.
  • Strep. bovis is Gram-positive and shows gamma-hemolysis on blood agar.
  • N. sicca is Gram-positive.
  • N. lactamica is Gram-negative.
  • M. catarrhalis is Gram-negative.

Physiological and Biochemical Tests

  • Staph. aureus, Micro. luteus, and Staph. saprophyticus produce yellow colonies on Mannitol Salt Agar, indicating mannitol fermentation. Staph. epidermidis produces pink colonies.
  • Strep. pyogenes, Strep. agalactiae, and E. faecalis grow on Mannitol Salt Agar but don't ferment mannitol. Strep. bovis does not grow on Mannitol Salt Agar.
  • Staph. aureus is catalase-positive (bubbles). Staph. epidermidis and Staph. saprophyticus also produce bubbles with catalase.
  • Staph. aureus produces Clots while Staph. epidermidis and Staph. saprophyticus do not.
  • Staph. aureus is novobiocin-sensitive, Staph. epidermidis is novobiocin-sensitive, and Staph. saprophyticus is novobiocin-resistant.
  • Staph. aureus is bacitracin-resistant and Nitrate positive.
  • Strep. pyogenes and Strep. agalactiae are CAMP negative, bile esculin negative, and PYR negative. Strep. agalactiae gives a positive Hippurate test. Strep. pyogenes gives a negative Hippurate test.
  • E. faecalis shows growth and blackening on bile esculin agar and is PYR positive (red).
  • Strep. bovis exhibits growth and blackening on bile esculin agar and is PYR negative.
  • N. sicca, N. lactamica, and M. catarrhalis grow on Modified Thayer-Martin (MTM) and Chocolate Agar (CA).
  • N. sicca, N. lactamica, and M. catarrhalis are oxidase-positive.

Additional Biochemical Tests

  • C. freundii is oxidase-negative, K/A with H2S on TSI.
  • E. coli is oxidase-negative, A/A with gas on TSI.
  • E. aerogenes is oxidase-negative, produces A/A and H2S with gas on TSI.
  • Proteus vulgaris is oxidase-negative, produces A/A and H2S with gas on TSI.
  • Providencia rettgari is oxidase-negative, K/A on TSI, is Indole and Citrate positive.
  • Salmonella typhimurium is oxidase-negative, K/A with H2S on TSI.
  • Shigella sonnei is oxidase-negative, K/A on TSI, is indole Negative
  • Proteus mirabilis is oxidase-negative, K/A H2S, Citrate and Urease positive.
  • Serratia marcescens is oxidase-negative, has A/A/Yellow (Yellow/Yellow) on TSI is Dnase -positive.
  • E. coli is Indole and SIM-positive, Voges-Proskauer and Citrate-Negative
  • E. aerogenes is Voges-Proskauer and Citrate -positive, indole negative.
  • Proteus vulgaris is SIM-positive (H2S production).
  • Serratia produces Lysine and is Positive with Growth on MacConkey agar.
  • E. Coli does not produce Lysine and gives a Positive on MacConkey agar.
  • E. Aerogenes produces Lysine and gives a Positive on MacConkey agar.
  • Proteus vulgaris produces Lysine and gives a Positive on MacConkey agar.
  • C. Freundii is negative for Lysine and MacConkey is Pink Growth
  • Salmonella Typhimurium is negative for Lysine and also MacConkey is negative (clear growth)
  • Shigella doesn't produce Lysine and grows negative on MacConkey agar.
  • Proteus mirabilis produces Lysine and produces a growth on MacConkey.
