Study Notes
Gene Transfer Overview
- Gene transfer involves moving genetic information between organisms
- This occurs through two main mechanisms: vertical and horizontal
- Vertical transfer is from parent to offspring; a normal process
- This includes mutations and genetic information passed down
- Horizontal transfer involves moving genetic material to non-offspring organisms
Mechanisms of Horizontal Gene Transfer
-
Horizontal gene transfer moves genetic information between individuals of the same or different species, not through reproduction
-
Key methods include transformation, conjugation, and transduction.
-
Transformation: Bacteria take up free DNA from their surroundings, usually from lysed cells. Natural competence exists in some species (like Bacillus) enabling direct uptake of foreign DNA
-
Conjugation: Direct contact between bacteria. The donor bacterium carries a fertility factor (F-factor) transferring DNA. This involves a process with four steps for effective transfer.
-
Transduction: Transfer of genetic material through viruses (bacteriophages) which infect bacteria. Pieces of the donor bacteria's DNA are packaged with phage DNA (often resulting in a recombinant cell).
Methods of Gene Transfer
- Physical methods: Examples include electroporation, thermal assisted gene transfer, and laser-assisted transfection.
- Chemical methods: Examples like Calcium Phosphate and DEAE-dextran-mediated procedures can make bacteria competent for DNA uptake
- Biological methods: Utilizing viruses as vectors (e.g., adenoviruses, retroviruses) for gene transfer is another strategy.
Electroporation
- Briefly applying a high-voltage electric pulse to cells creating transient pores in the plasma membrane for DNA entry. This technique is often used for introducing DNA.
Heat Shock Transformation
- Calcium chloride is commonly used to make E. coli competent for uptake of DNA. A brief heat shock makes the cell membrane permeable. This technique is also associated with lower competency compared to electroporation
Chemical Transformation
- Heat shock involving cells in a chemical solution (e.g. calcium chloride) is performed for a specific time, which is optimized for the bacteria used in the procedure.
Applications of Gene Transfer
- Recombinant antibody production
- Vaccine development
- Gene therapy
- Producing transgenic animals
- Discovering new genes
- Creating improved crops
- Developing pest/herbicide strategies.
