Dantrolene
What is Dantrolene?
- A medication used to treat malignant hyperthermia, a life-threatening medical emergency
- Also used to treat muscle spasms, spasticity, and other muscle disorders
Mechanism of Action
- Works by reducing the release of calcium from the sarcoplasmic reticulum, which leads to muscle relaxation
- Inhibits the ryanodine receptor, a key player in muscle contraction
Indications
- Malignant hyperthermia
- Muscle spasms
- Spasticity
- Neuroleptic malignant syndrome
- Heat stroke
Administration
- Available in oral and intravenous forms
- Oral form used for chronic treatment of muscle spasms and spasticity
- IV form used for malignant hyperthermia and other acute conditions
Side Effects
- Drowsiness
- Dizziness
- Diarrhea
- Fatigue
- Muscle weakness
Contraindications
- Active hepatic disease
- Impaired pulmonary function
- Cardiac disease
- Pregnancy and lactation (category C)
Special Considerations
- Can cause hepatotoxicity, monitor liver function regularly
- May interact with other medications, such as MAOIs and SSRIs
- Use with caution in patients with impaired renal function
