- Bacteria are classified according to their immunologic and genetic characteristics
Gram Stain
- Gram stain is the most popular means of visualizing bacteria
- It groups bacteria into 2 categories: Gram-Positive bacteria and Gram-Negative bacteria
- Gram stain helps to visualize the shape of bacteria
- The difference in stains is due to differences in the cell wall of bacteria
Gram-Positive Bacteria
- Appear blue under Gram stain
- Have a thicker peptidoglycan layer
Gram-Negative Bacteria
- Appear red under Gram stain
- Have a thinner peptidoglycan layer
Bacterial Cell Structure
- Bacteria have a cytoplasmic membrane that surrounds the cytoplasm
- They also have a peptidoglycan layer (also known as the cell wall) that surrounds the cytoplasmic membrane
- The peptidoglycan layer is composed of repeating disaccharide units with amino acid side chains
- The monomers are held together by the enzyme transpeptidase, located on the inner surface of the cytoplasmic membrane
