  • Bacteria are classified according to their immunologic and genetic characteristics

Gram Stain

  • Gram stain is the most popular means of visualizing bacteria
  • It groups bacteria into 2 categories: Gram-Positive bacteria and Gram-Negative bacteria
  • Gram stain helps to visualize the shape of bacteria
  • The difference in stains is due to differences in the cell wall of bacteria

Gram-Positive Bacteria

  • Appear blue under Gram stain
  • Have a thicker peptidoglycan layer

Gram-Negative Bacteria

  • Appear red under Gram stain
  • Have a thinner peptidoglycan layer

Bacterial Cell Structure

  • Bacteria have a cytoplasmic membrane that surrounds the cytoplasm
  • They also have a peptidoglycan layer (also known as the cell wall) that surrounds the cytoplasmic membrane
  • The peptidoglycan layer is composed of repeating disaccharide units with amino acid side chains
  • The monomers are held together by the enzyme transpeptidase, located on the inner surface of the cytoplasmic membrane
