HEMOGLOBIN ESTIMATION BY SAHLI'S METHOD REQUIREMENTS Sahli's hemoglobinometer N/10 hydrochloric acid (HCl) & Distilled water HEMOGLOBIN ESTIMATION BY SAHLI'S METHOD REQUIREMENTS Lancet Alcohol swab / Alcohol pad Dropper SAHLI'S HEMOGLOBINOMETER Sahli's hemoglobinometer consists of: Comparator - It has a pair of standard nonfading coloured glass. Hemoglobinometer tube - graduated in grams per 100ml on one side and in percentage on the other side. The readings on the hemoglobin tube is from 2 to 22gm% The graduation on the tube is in the division of 0.2 For example it starts as 2.0 , 2.2 , 2.4 , 2.6 , 2.8 , 3.0 ……… SAHLI'S HEMOGLOBINOMETER Sahli's hemoglobinometer consists of: Pipette with 20 cu mm capacity to suck blood Glass rod to stir Principle of Sahli's hemoglobin estimation The blood is mixed with N/10 hydrochloric acid and the hemoglobin released from the hemolysed red blood cells combines with the acid to form acid hematin. The mixture is then diluted with distilled water Finally the hemoglobin content is determined by matching the solution against a standard (permanent) non-fading coloured glass. PROCEDURE OF HEMOGLOBIN ESTIMATION 1) Place N/10 Hydrochloric acid in the hemoglobin tube up to the mark 2 gm% (yellow marking in the Hemoglobin tube) 2) The finger is sterilized with an alcohol swab and pricked with a lancet. PROCEDURE OF HEMOGLOBIN ESTIMATION 3) Draw the blood up to 20cu mm or 20μl mark in the hemoglobin pipette (avoid air bubbles in the blood column and wipe off the excess blood from tip of the pipette) PROCEDURE OF HEMOGLOBIN ESTIMATION 4) Dip the pipette into the hemoglobin tube and empty the blood into it 5) It is mixed with the glass rod and left free for about 5 minutes 6) The mixture would turn brown in colour due to the formation of acid hematin Hemoglobin + HCl = Acid hematin PROCEDURE OF HEMOGLOBIN ESTIMATION 7) Dilute the blood with distilled water drop by drop till the colour of the solution matches with the colour of the standard. PROCEDURE OF HEMOGLOBIN ESTIMATION 8) The hemoglobin content can be read off directly (lower meniscus) in grams/100 ml or grams/dl NORMAL RANGE OF HEMOGLOBIN Normal value for men = 14 to 18 grams/100 ml Normal value for women = 12 to 16 grams/100 ml.
