Flashcards

Lectin Pathway

Initiated by mannan-binding lectin on microbes, activating C4 and C2.

C1-like complex

A complex similar to C1qrs, initiates this cascade upon lectin binding.

C4

Cleaved into C4a and C4b during the Lectin and Classical pathways.

C3 Convertase (Lectin/Classical)

Formed by C4b and C2a, cleaves C3 into C3a and C3b

C5 Convertase (Lectin/Classical)

Formed by C4b2a and C3b, cleaves C5 into C5a and C5b.

Study Notes

  • There are 3 complement pathways: classical, lectin and alternative

Lectin Pathway

  • Carbohydrate-containing mannose binds with MBL (mannose-binding lectin)
  • C4 is cleaved into C4a and C4b
  • C2 is cleaved into C2a and C2b
  • C4b and C2a combine to form C4b2a (C3 convertase)
  • C3 convertase activates C3
  • C3 is cleaved into C3a and C3b
  • C4b2a and C3b combine to form C4b2a3b (C5 convertase)

Classical Pathway

  • Antigen-antibody complex activates C1-like complex
  • C4 is cleaved into C4a and C4b
  • C2 is cleaved into C2a and C2b
  • C4b and C2a combine to form C4b2a (C3 convertase)
  • C3 convertase activates C3
  • C3 is cleaved into C3a and C3b
  • C3b leads to the polymerization of C9
  • C5 convertase cleaves C5 into C5a and C5b
  • C5b, C6, and C7 combine to form C5b6C7 complex
  • C5b6C7 binds to C8
  • C5b6C78 leads to lysis of pathogen

Alternative Pathway

  • Spontaneous hydrolysis occurs with C3 to generate C3(H2O)
  • Factor B binds with C3(H2O) to form C3(H2O)B
  • Factor D cleaves Factor B into Ba and Bb
  • C3(H2O) and Bb combine to form C3(H2O)Bb (fluid phase C3 convertase)
  • C3 convertase activates C3
  • C3 is cleaved into C3a and C3b
  • Properdin (P) binds and stabilizes C3bBb to form C3bBb3b (C5 convertase)
  • C5 convertase cleaves C5 into C5a and C5b
